A quick summary of the replies I got offlist:
For software the original bind path lives here:
http://www.caraytech.com/geodns/
and a couple of people recommend the powerdns geobackend module:
http://wiki.powerdns.com/trac/browser/trunk/pdns/modules/geobackend/README
For vendors I got recommendations for:
Dynect from Dyn Inc - http://dyn.com/dynect
Global Traffic Management from Akamai - http://www.akamai.com/html/technology/products/gtm.html
All four of the above appear to be used by large and serious sites.
